On the cover, isolation and proteomic characterization of basolaterally targeted exocytic vesicles has proven to be a formidable challenge. The authors have exploited the myristoylation-dependence of Naked2 to achieve fusion of Naked2-associated, TGF-α- containing exocytic vesicles at the basolateral surface of polarized epithelial cells. They biochemically enriched myristoylation-deficient G2A Naked2 vesicles that were trapped in the cytoplasm, performed FAVS, and then carried out LC/LC-MS/MS analysis. Three hundred and eighty nine proteins were found in three independent experiments, and, as an initial validation step, six of these (including Na+/K+-ATPase α1) were present in wild-type Naked2 vesicles.
